摘要
目的观察果糖二磷酸镁(FDP-M)对心肌缺血-再灌注损伤过程中血清中CK,LDH,Mg2+浓度、心肌组织内SOD,MDA浓度的影响,探讨果糖二磷酸镁心肌保护作用的机理及其剂量依赖型。方法用整体大鼠急性心肌缺血再灌注损伤模型,结扎冠脉左室前降支40min后,再灌注40min,应用比色法测定血清中CK,LDH,Mg2+含量,采用羟胺法测定心肌组织中SOD含量,采用硫代巴比妥钠(TAB)比色法测定心肌组织内MDA含量。结果与假手术组相比,缺血再灌注损伤组血中CK,LDH浓度显著升高,Mg2+浓度显著降低,组织内MDA含量明显升高,SOD含量明显降低。与缺血损伤组相比,果糖二磷酸镁三个不同剂量保护组血中CK,LDH浓度明显降低,Mg2+浓度明显升高,组织内MDA含量明显降低,SOD含量明显升高,差异具有显著性(P<0.05或P<0.01),此作用均可见明显剂量依赖性。结论果糖二磷酸镁对心肌缺血再灌注损伤有保护作用,并呈明显的剂量依赖型,其保护作用机制与增加组织内SOD含量,减少血中CK,LDH浓度,减少组织内MDA含量有关。
Objective This paper was designed to observe the concentration change of CK, LDH, Mg^2+ in the blood serum and SOD, MDA in the myocardial to investigate the cardioprotective effect of Magnesium Fructose-1,6-diphosphate during myocardial ischemia reperfusion injury in anesthetized rats. Methods We adopted the model of myocardial ischemia reperfusion injury in anesthetized rats, determined the concentration of CK, LDH and Mg^2+ in blood serum, and we determined the contents of SOD and MDA in myocardium in the same time. Results The concentration of Mg^2+ and SOD were decreased and the concentration of CK, LDH and MDA were increased in the ischemia reperfusion group compared with the sham, and concentration of Mg^2+ and SOD were significantly increased and CK, LDH, MDA contents were decreased in the three groups of Magnesium Fructose-1,6-diphosphate compared with the ischemia reperfusion group. The dosage mentioned above. Conclusions Magnesium Fructose-1,6-dependence was observed of the effect diphosphate has protective effects on myocardial ischemia reperfusion injury. The protective effect may be related to increasing concentration of Mg^2+ and SOD, relieving CK, LDH,MDA and antilipid peroxidation.
