摘要
目的探讨转化生长因子(TGF-β1)多克隆抗体作为激光泪道成形术后辅助用药的可行性。方法使用相同能量倍频-掺钕钇铝石榴石(KTP)激光对兔泪小管进行泪道成形术,分别使用TGF-β1多克隆抗体与丝裂霉素作为激光泪道成形术后的辅助用药,观察其治疗效果。通过组织病理学方法观察治疗后的组织修复过程,通过免疫组织化学方法检测TGF-β1,TGF-β2在修复过程中的分布与表达,通过苦味酸天狼猩红特殊染色法检测泪小管组织胶原纤维含量,采用图文分析系统对结果进行半定量分析。结果TGF-β1在丝裂霉素组及抗体组术后1及7 d时明显低于对照组,术后1个月含量升高,高于对照组;TGF-β2在丝裂霉素组及抗体组术后1 d低于对照组,术后7 d至1个月含量显著升高;丝裂霉素组胶原纤维的含量在7 d及1个月时显著升高,抗体组1 d^1周时达到高峰。丝裂霉素组与抗体组胶原纤维含量从术后7 d起均低于对照组,1个月时差异有显著性。结论丝裂霉素与TGF-β1多克隆抗体对TGF-β1及TGF-β2的分布与表达均能产生一定的影响,对胶原纤维的增生有抑制作用,从而减轻泪小管的阻塞程度。因此,TGF-β1多克隆抗体有可能成为激光泪道成形术后的辅助药物。
Objective To assess the feasibility of using anti-TGF-β1 polyelonal antibody as an assistant therapy after laser canalicular plastic operation. Methods KTP/YAG laser canalicular plastic surgery was performed at fixed power with combination of injection of anti-TGF-β1 polyelonal antibody or mitomyein-C (MMC) to rabit's lacrimal canaliculus. And those operated without any injection were taken as the control group. The histopathalogy of lacrimal canalieulus was studied by means of HE staining. Immunohistochemieal technique was used to detect the distribution and expression of TGF-β1 and TGF-β2. Picrosirius method was used to detect the collagen content in canalicular tissue. The half-quantitative analysis of the distribution and expression of TGF-β1, TGF-β2 and collagen were assessed by HMIAS-2000 color pathologic analysis system. Results The expression of TGF-β1 and TGF-β2 was significantly suppressed on day 1 to week 1 after operation in the group of anti-TGF-β1 antibody or mitomycin-C. The content of collagen in group of anti-TGF-β1 antibody or mitomycin-C was lower than that of control group after week 1. Conclusion Mitomycin-C and anti-TGF-β1 antibody can influence the distribution and expression of TGF-β1 and TGF-β2, and in a certain degree, inhibit the hyperplasia of collagen fibers, thus reducing the blocking of canaliculus. Therefore, anti-TGF-β1 antibody has a potential as a new assistant therapy for reducing post-surgical scarring.
