摘要
目的探讨老龄大鼠海马结构trkB mRNA表达的变化,同时对比观察人参皂苷对其改变的影响,为人参皂苷抗脑衰老及提高老龄动物学习记忆能力提供可靠的实验依据。方法选用W istar♀大鼠24只,随机分为青年组、老龄组、给药组(从第17个月始饲以人参皂苷至27个月龄)。用原位杂交组化方法(ISH)结合图像分析技术对海马结构TrkBmRNA进行定性、定量分析。结果老龄组CA3、CA1区及齿状回神经元TrkBmRNA表达明显下调,含量分别比青年组下降了24.2%、13.4%和50.6%(P<0.01);给药组CA3区和齿状回TrkB mRNA表达上调,含量分别比老龄组增加了12.7%(P<0.05)和17.9%(P<0.01),但给药组CA1区表达改变不明显(P>0.05)。结论老龄大鼠海马结构3个脑区的TrkB mRNA表达明显下调,而人参皂苷具有上调CA3区和齿状回TrkB mRNA表达的作用。
Aim To investigate the age-related changes of TrkB mRNA in hippocampal formation of aged rats, and the effects of ginsenosides(GS) ,and provide reliable experimental evidence for anti-aging. Methods 24 Wistar female rats were randomly divided into 3 groups: Young, aged and GS group (fed with GS from 17 to 27 months). In situ hybridization (ISH) method was applied into qualifying and quantitating the TrkB mRNA expression in hippocampal formation. Results TrkB mRNA expression in aged CA3, CA1 and dentate gyms decreased by 24. 2%, 13.4% and 50. 6% (P 〈 0. 01 ) respectively compared with the young group, while TrkB mRNA expression in CA3 and den- tate gyrus of GS group increased by 12.7% (P 〈0. 05) and 17. 9% (P 〈0. 01 ) compared with aged group, respectively. But there is no significant change in the CA1 in GS group. Conclusion TrkB mRNA in CA1, CA3 and dentate gyrus neurons of the aged hippocampal formation declined markedly. However, GS could improved TrkB mRNA expression in CA3 and dentate gyrus in hippocampal formation.
