摘要
目的:克隆测序新分离呼肠病毒的S1全长基因组片段,并对其进行序列分析。方法:从接种呼肠病毒的L929细胞中提取病毒基因组,用改进的单引物扩增技术获得S1片段的全长cDNA克隆井测定其全序列。结果:新分离呼肠病毒的S1片段长1437bp,其核苷酸序列与已知的1—3型呼肠病毒标准株的同源性分另1为59%,61%和48%;推测的cr1序列与标准株的同源性分别为52%,60%和26%。结论:新分离呼肠病毒的S1片段与目前已知的分离株相比有很大差异,有可能是2型呼肠病毒的一个独立分支。
Objective: To clone, sequence and analyze genome segment 1 (S1) of new isolated reovirus strains. Methods: Virus genome was extracted from L929 cell infected with the isolate. Full-length eDNA clone of S1 segment was obtained by improved single primer amplification technique. Nueleotide and amino acid sequences were analyzed using free computer softwares. Results: The total nueleotide length of S1 segment was 1 437 bp. Nueleotide homology with 3 prototypie strains T1L, T2J and T3D was 59% ,61%, and 48%, respectively. Amino acid homology of deduced sigmal protein with 3 prototypie strains was 52%, 60%, and 26%, respectively. Conclusion: S1 segment of new isolated reovirus is remarkably different from that of other known strains, and it might belong to an independent branch of serotype 2 reovirus in evolutionary tree.
