摘要
目的观察外源性磷酸肌酸、左旋精氨酸对大鼠供心保存效果。方法40只健康SD大鼠随机分为4组,移植前供心保存480 min,对照组:供心用4℃的St.Thomas液保存;1组:保存液中添加磷酸肌酸(CP);2组:保存液中添加左旋精氨酸(L-arg);3组:保存液中添加磷酸肌酸及左旋精氨酸。保存后进行颈部异位心脏移植术,并观察供心复跳情况。24 h后处死受体动物,切除移植心脏,测心肌组织中ATP酶、超氧化物歧化酶(SOD)、丙二醛(MDA)含量,电镜观察心肌超微结构改变。结果实验组移植心脏自动复跳率高于对照组。3组心肌组织中ATP酶、SOD含量均高于对照组及1组2、组(P<0.05),而MDA含量则低于对照组及1组、2组(P<0.05)。心肌超微结构损伤较对照组与组1、组2轻。结论外源性磷酸肌酸、左旋精氨酸能明显增强保存液对心脏的保护作用。
Objective To investigate the effects of creatine phosphate (CP) and L - arginine ( L - arg) on the preservation of donor heart in rats. Methods Forty rats were randomly allocated into 4 groups to get the hearts preserved for 480 min with 4℃ cardioplegic solution (control group), with cardioplegic solution plus CP (group 1 ), with cardioplegic plus L- arg (group 2) and with cardioplegic plus CP and L - arg (group 3). The donor heart was transplanted into the neck of recipient rat. Left ventricular tissue was removed 24 h after transplantation to determine the contents of SOD, ATPase and MDA, and to study the myocardial ultrastrueture. Results Arrhythmia was much less frequent in group 3 than in control group, group 1 and group 2. The contents of SOD and ATPase were higher, but the content of MDA was lower in group 3 than in the other 3 groups. The damages in myocardial ultrastrueture were also less severe in group 3. Conclusion Exogenous CP and L -arg could significandy enhance the eardioplegia effects.
