摘要
研究rt-PA工程细胞在小型填充床反应器中连续高密度培养的过程,以及在连续灌注培养条件下rt-PA表达水平、葡萄糖的消耗率、乳酸和氨与rt-PA表达水平之间的关系以探索该工程细胞培养的最佳条件。在不中断培养的情况取样作细胞计数、rt-PA和代谢物的检测。结果:rt-PA工程细胞在小型填充床反应器中最高细胞数可达3.8×109细胞,最高rt-PA浓度达12mg/L结论:这种新型的小型填充床反应器是操作方便、利用价值高的研究设备。
PURPOSE To study the process for high density culture of rt-PA engineering cells and the relationship between the expression of rt-PA and consumption rate of glucose, lactate and ammnia productivity. METHODS The complete procedure of sampling and counting cells were established under the continuous culture condition. RESULTS The cells grown in the mini packed-bed bioreactor was 3.8 × 109 cells and the highest concentration of t-PA was 12 mg/L CONCLUSIONS The newly-designed mini packed-bed bioreactor is an easy and high efficient bioreactor for cell culture.
