摘要
目的从大蒜中定向分离具有拮抗脓毒症作用的物质。方法利用生物传感器等技术,对大蒜中活性组分进行定向分离;观察活性组分对LPS刺激RAW264.7细胞分泌炎症介质的抑制作用;观察活性组分对LPS攻击小鼠的保护作用。结果从大蒜中分离出4个活性组分,其中以2组分(DS2)的活性最高。DS2对LPS刺激RAW264.7细胞分泌炎症介质具有显著的抑制作用,对致死剂量LPS攻击的小鼠具有显著的保护作用。结论从大蒜中定向分离出的DS2组分具有显著的拮抗LPS活性。
Objective To obtain the active components from allium sativum L. Methods The active fractions were isolated by biosensor technique,and theirs inhibition on TNF-α release from RAW 264.7 cells induced by LPS were observed. The protection of the fraction on mice from lethal challenge of LPS were observed in vivo and in vitro. Results The active fraction named as DS2 with the best affinity for lipid A was separated. DS2 markedly inhibited TNF-α release from RAW264.7 cells induced by LPS, and protected mice from lethal challenge of LPS. Conclusions The DS2 has significant activity against sepsis.
