维甲酸对胃癌细胞生长抑制及凋亡的诱导作用 被引量:12

9-cis-RA induced apoptosis and inhibited proliferation in human gastric cancer cell line MGC80-3

摘要 目的:研究9-顺-维甲酸(9-cis-RA)对人胃低分化黏液腺癌细胞(MGC80-3)的作用。方法:MTT比色法观察9-cis-RA对MGC80-3细胞株的生长抑制作用,倒置显微镜观察细胞形态变化,Hoechst33342/PI双荧光染色和DNA凝胶电泳技术分析细胞凋亡,流式细胞仪检测细胞周期的改变。结果:9-cis-RA可抑制MGC80-3细胞生长,半数抑制浓度(IC50)为8·07×10-6mol/L;显微镜观察可见细胞凋亡特征性改变;10-5mol/L浓度的9-cis-RA药物作用后96h可明显诱导MGC80-3细胞凋亡,琼脂糖凝胶电泳检测到DNA梯带;流式细胞仪检测肿瘤细胞出现典型亚二倍体凋亡小峰,G0/G1期细胞百分率明显升高,由对照组的(61·5±2·2)%增加到(74·3±4·7)%,F=149·795,P=0·000;同时处于S期的细胞数减少,由对照组的(26·3±1·3)%减少到(17·2±1·4)%,F=143·936,P=0·000。细胞周期出现G1期阻滞。结论:9-cis-RA对MGC80-3细胞有明显的生长抑制和诱导凋亡作用。 OBJECTIVE: To study the effect of 9-cis-RA on the human gastric cancer cell line MGC80-3. METHODS: MGC80-3 was treated with 9-cis-RA at different concentrations. The growth was analysed by MTT assay and the cells were observed with an invert microscope in shape and with a fluorescence microscope in apoptosis dyed by Hoechst33342/PI and agarose gel electrophoresis. The effect of 9-cis-RA on the cycle of tumor cells was analyzed by flow cytometry. RESULTS: Treatment of MGC80-3 cells with 9-cis-RA inhibited cell growth. The IC50 value of 9-cis-RA for MGC80-3 cells was 8.07× 10^-6mol/L. A sign of apoptosis was observed in morphological changes with a microscope. Apoptosis of MGC80-3 cells was induced by 9-cis-RA at concentration of 10^-5mol/L at 96 hours. After 96 hours treated with 9-cis-RA, some apoptotic cells were detected and a typical subdiploid peak was observed by flow cytometry, and the percentage of G0/G1 phage cells increased from (61.5 ± 2.2)% to (74.3±4.7)%, F=149.795, P=0.000, and decreased of S phage cells from (26.3± 1.3)% to (17.2 ± 1.4)%, F= 143. 936,P=0.000,and G1 phage of the cell cycle was arrested. CONCLUSION: 9-cis-RA could efficiently induce apoptisis of MGC80-3 cells.
