摘要
目的观察脂蟾毒配基对人宫颈癌Hela裸鼠移植瘤生长的抑制作用。方法于给药前、给药期间(15d)及停药期间(6d)每3d测量肿瘤的长径及短径,并计算近似瘤体积,停药第6天取瘤称重并进行病理学观察。结果①高剂量脂蟾毒配基(15mg/kg)有较强的急性神经毒性。②脂蟾毒配基对荷瘤鼠体重无明显影响。③高剂量脂蟾毒配基(15mg/kg)对人宫颈癌Hela裸鼠移植瘤生长有明显的抑制作用,停药后没有明显的后续抑制作用,15mg/kg的脂蟾毒配基与5mg/kg平阳霉素对肿瘤增长的抑瘤强度相当。④脂蟾毒配基未发现明显的近期毒性。结论脂蟾毒配基对裸鼠的Hela宫颈癌移植瘤有明显的抑瘤作用,脂蟾毒配基是中药蟾酥抗癌的重要成分,但也发现该化合物有明显的急性神经毒性,进一步开发需要慎重。
Objective To observe the inhibitory effect of resibufogenin (RBG) on transplantation tumor of human cervical cancer Hela cells in rude mice. Method The short and long diameter of the tumor was measured every 3 days before treatment, in treatment period ( 15 days) and rest period (6 days) respectively, and the approximate volume of tumor was calculated. On the 6th day after stopping treatment the tumor was weighed and observed pathologically. Results ①The large-dose of RBG ( 15 mg/kg) had a strong acute neurotoxicity. mouse. ② RBG had no obvious influence on weight of tumor-beating mouse. ③The large-dose of RBG ( 15 mg/kg) had an obvious inhibitory effect on transplantation tumor of human cervical cancer in nude mouse, but no follow-up effect after stopping treatment. RBG indose of 15 mg/kg and Pingyangmycin in dose of 5 mg/kg were about equal in tumor-inhibitory effect.④The short-term neurotoxicity of RBG was not observed. Conclusion RBG has a remarkable tumor-inhibitory effect on transplantation tumor growth of human cervical cancer Hela cells in nude mice. RBG is an important principle with anti-cancer effect of Chinese herbal drug, vallicepobufagin, but it also has acute neurotoxicity. The further development of RBG should be handled with great care.
