滋养肝肾、活血化瘀中药对实验性糖尿病大鼠视网膜超微结构的影响

The influence of preparation of liver and kidney hourishing,blood flow-activcting and stasis-re-moving traditional Chinese medicine on the ultrastructure in retina of experimental diabetic rats

目的：观察６个月病程四氧嘧啶性糖尿病大鼠视网膜病理改变以及滋养肝肾、活血化瘀中药对实验性糖尿病大鼠视网膜超微结构的影响。方法：将６０只四氧嘧啶所致的糖尿病大鼠随机分为４组：中药高剂量组、中药低剂量组、达美康对照组及生理盐水对照组，并设正常对照组。每日灌药１次，６个月后每组随机处死２只大鼠取视网膜制片作电镜观察。结果：生理盐水对照组糖尿病大鼠视网膜毛细血管内皮细胞、周细胞核异染色质聚集、靠边，胞质内线粒体肿胀变性；基底膜结节样增厚；毛细血管壁断裂；视杆外节变性、破坏；色素上皮微绒毛结构紊乱。中药治疗组糖尿病大鼠视网膜超微结构病理损害明显轻于生理盐水对照组。结论：６个月病程时四氧嘧啶性糖尿病大鼠已具有糖尿病性视网膜病变的早期病理改变。滋养肝肾、活血化瘀中药对早期糖尿病性视网膜病变具有较好的防治作用。

OBJECTIVE:This studywas designed to investigate the pathological change inretina of alloxan-diabetic rats at the duration of 6 months and the influence of preparation of liverand kidney hourishing, blood flow-activeting and stasis-removing traditional Chinese medicineon the ultrastructure in the retina METIIODS:Sixty rats with diabetic induced by alloxan were di-vided randomly into 4 groups:large dose preparation of the traditional Chinese medicine group,small dose preparation group,diamiand group and normal saline groups.Normal control groupwas also set.After 6 months of administration of the drug once daily,2 randomly selected formeach group,were killed and the sectons of the retine were made and examined with transmissionelectron microscope, RESULTS:In the normal saline group the pathological changes of the retinaincluded aggregation and margination of heterochromatins in the、nuclear of endothelial cells andpericytes,swelling and deterioration of mitochondria in cytoplasm,nodular thickening basementmembranes and breaking of capillary in some plaees,degeneration of photosensory cells outer seg-ment and disturbanee of microvilli of pigment epithelial cells,but the pathological damages of theultrastructures in the retina from the traditional Chinese medicine group were abated significantly,CONCLUSIONS: At the duration of 6 months there have been early diabetic retinopathy changesin retina of alloxan-diabetic rats.The preparation of tradilional Chinese medicine had effect ofpreventing and treating diabetic retinopathy at early stage.
