摘要
利用形态观察法、台盼蓝排染法、集落形成法及姬姆萨染色法研究了从甘肃产拟缺香茶菜(Isodon excisoides(Sun ex C.H.Hu)C.Y.Wu et H.W.Li)中分离得到的二萜化合物Wangzaozin A对人胃腺癌细胞SGC-7901的增殖抑制作用及致死效应.结果表明,二萜化合物Wangzaozin A对SGC-7901细胞的生长状况有显著影响,具体表现为低浓度(≤4μmol.L-1)条件下抑制细胞增殖,高浓度(≥8μmol.L-1)条件下致细胞死亡,且具有时间-剂量依赖效应;姬姆萨染色显示,在高浓度条件下,Wangzaozin A能诱导人胃腺癌细胞SGC-7901发生凋亡.
The effect of Wangzaozin A, an ent-kaurane diterpene isolated from Isodon excisoides (Sun ex C. H. Hu) C. Y. Wu et H. W. Li, on the proliferation of human cancer SGC-7901 cells is investigated by morphological observation, typanblue exclusion method, colony-forming test and Giemsa staining. The results exhibite that Wangzaozin A markedly inhibite the proliferation of SGC- 7901 cells at low concentrations(≤4 μmol·L^-1 ) while made cells death at mass concentrations(≥8μmol·L^-1 ), and both in the time-and dose-dependent manners. The morphology of SGC-7901 cell nuclei observed by Giemsa staining, show morphological characteristics of apoptotic cells, Wangzaozin A can induce apoptosis in SGC-7901 cells.
