摘要
目的探讨应用5-溴-2-脱氧尿苷(5-bromo-2-deoxy-uridine,BrdU)标记检测大鼠生精细胞体外的分化、发育情况。方法采用BrdU体内标记生精细胞的DNA,免疫细胞化学法跟踪观察大鼠生精细胞在体外培养中的分化、发育情况。结果培养前在体内标记BrdU的细胞为初级精母细胞,经培养后在BrdU标记的细胞中出现早期精子细胞。结论大鼠生精细胞体外培养过程中,应用BrdU标记能检测生精细胞的分化、发育情况。
[Objective] To investigate differentiation and development of rat spermatogenic cells cultured in vitro by application of 5-bromo-2-Deoxyufidine(BrdU)-labeled immunoeyto-chemistry. [Methods] Rats were injected with BrdU 9 days before experiment in order to label the spermatogenic cells' DNA in vivo. The BrdU-labeled immunocytoehemistry was used to observe spermatogenic cell differentiation and development in vitro. [Results] BrdU-labeled immunocyto-chernistry showed that BrdU-labeled early phase spermatids were observed after the labeling primary spermatocytes cultured in vitro. [Conclusion] BrdU-labeled inununoeytochemistry can be used to detect the differentiation and development of spermatogenie cells culture in vitro.
