摘要 目的筛选复方柴胡口服液中橙皮苷的水煎煮提取工艺,比较乙醇沉淀和壳聚糖澄清剂的澄清工艺。方法采用L9(34)正交设计,考察加水量、煎煮时间、煎煮次数3个因素,以青皮中有效成分橙皮苷作为评价指标,采用高效液相色谱法测定橙皮苷的含量。结果煎煮次数对橙皮苷的含量有显著性影响,溶媒用量对橙皮苷的提取有较大的影响;壳聚糖吸附澄清法与醇沉法均能使药液澄清。结论水提最佳工艺确定为加水18倍量,每次1.5 h,煎煮3次;壳聚糖澄清剂可以替代乙醇沉淀;优选得到的提取工艺稳定可行。 Objective:To optimize the extraction technics of hesperidin from compound Chaihu oral liquid and compare the sedimentation process by ethanol and chitosan. Methods The orthogonal-test method was adopted to study the effects of 3 factors: the volume of water, the duration and times of extraction. Determination of hesperidin by HPLC was used as appraisement index. Results The number of extraction and the amount of water had significant influence on the content of hesperidin. Both processes by chitosan and ethanol had clearing action. Conclusion The optimum technics for extracting hesperidin from compound Chaihu oral liquid was as follows: the volume of water was 18 times larger than raw herb, the optimum extracting times was 3 and the duration of extraction was 1.5 h each time. Chitosan process could substitute ethanol process, and the extraction technics was stable and feasible.
