血小板第4因子对放射损伤小鼠骨髓基质细胞周期的影响

The effect of platelet factor 4 on cell cycle of bone marrow stromal cells in radiated mice

摘要 目的探讨血小板第4因子(plateletfactor4,PF4)对5·0Gyγ射线全身照射小鼠的骨髓基质细胞(bonemarrowstromalcells,BMSCs)的保护作用,进一步探讨PF4对造血的辐射防护机制。方法30只雄性小鼠随机分为3组:①放射组,②PF4保护组,③对照组。小鼠照射前分别于26和20h腹腔内注射PF4,每次剂量50μg/kg。于照射后3d取骨髓细胞体外培养,分别计数培养后3、7和14d的骨髓基质细胞集落(CFU-F);在培养后10d流式细胞仪检测细胞周期。结果3组中,照射组3d的CFU-F数量与PF4保护组差异无统计学意义,7和14d的CFU-F数量PF4保护组较照射组明显增加。流式细胞仪检测结果表明3组中照射组G0+G1期细胞明显高于其余两组,S,G2+M期细胞明显低于其余两组。结论PF4对照射小鼠的骨髓基质细胞有保护作用,促进造血重建。 Objective To investigate the protective effects of platelet factor 4 (PF4) on bone marrow stromal cells (BMSCs)in the irradiated mice and explore its mechanism. Methods Thirty male mice were randomly divided into three groups; mice in one of the three groups were exposed to whole-body irradiation of 5.0 Gy ^60Co gamma rays. The mice were intraperitoneally injected with PF4 (50μg/kg) at 26 h and 20 h before the irradiation. The CFU-F was counted at 3 d,7 d, and 14 d after the cell culture, respectively. Cell cycle of BMSCs cultured in vitro after irradiation, were analyzed by FACS at 10 d since the culture. Results Compared with the group protected by PF4, the difference of number of CFU-F at 3 d in irradiation group was not statistically significant. But the difference at 7 d or 14 d was statistically significant. The cell percentage of Go + G1 phase in the irradiation group increased, while the percentage of S phase and G2 + M phase decreased. Conclusion PF4 could nrotect BMSC in the irradiated mice and promote recoverv of hematopoiesis.
