快速溶剂萃取反相高效液相色谱法测定青蒿中的青蒿素 被引量:22

摘要 报道了一种应用快速溶剂萃取、经衍生化处理后用RP-HPLC测定青蒿药材中青蒿素的方法。青蒿样品用无水乙醇萃取剂在90℃,12.6MPa压力下萃取10min,用碱衍生化后,色谱测定,回收率在95.3%-101.2%。色谱分析条件采用Diamonsil C18色谱柱(250mm×4.6mm,5μm),甲醇-0.02mol/L乙酸铵为流动相,梯度:甲醇体积分数20%-35%,5min;35%-98%,5min;98%,6min,检测波长260nm;流速:1.0mL/min;柱温:25℃。结果表明该法准确、重现性好,可以为青蒿质量标准的制订提供科学依据。 procedure tracted by A rapid pressurized-solvent extraction, followed a derivation reaction and reversed-phase HPLC analysis has been developed to determine the content of artemisinine in Artemisia annua L.. Artemisinine was exanhydrous ethanol for 10 min at temperature 90℃ and pressure 12.6 MPa. Extracts were derived by alkali and then analyzed by HPLC. The recovery was from 95.3% to 101.2%. A Diamonsil C18 column was used with a mixture of methanol - 0.02 mol/L ammonium acetate solution as the mobile phase, the detection wavelength is at 260 nm. The flow rate was set at 1.0 mL/min, and the column temperature was 25℃. The results showed that the method developed is accurate , rapid and repeatable.
