摘要
目的:探讨β-羧乙基锗倍半氧化物(Ge-132)在体内清除自由基、抗氧化作用。方法:下列药物作用于烈酒造成氧应激态的大鼠,观察其肝线粒体超氧化物歧化酶活性和肝脏的病理变化,并与维生素C进行了比较。Wistar大鼠随机分为4组。正常对照(A),烈酒(B),烈酒+维生素C(C),烈酒+Ge-132(D)。各组大鼠常规饲喂。每种药的给药方法如下:烈酒(含体积分数为55%乙醇),5g/(kgd),从第8天开始ig共2周;维生素C200mg/(kgd),从第1天开始sc共3周;Ge-132200mg/(kgd),从第1天开始ig共3周。结果:肝线粒体铜锌超氧化物歧化酶活性,烈酒组与对照组比较,其活性下降(P<0.05),烈酒+Ge-132和烈酒+维生素C组均与烈酒组比较,其活性有上升趋势。肝脂肪变性在烈酒+Ge-132组较烈酒组为轻,烈酒+维生素C组病变不明显。结论:Ge-132在一定程度上保护肝线粒体铜锌超氧化物歧化酶活性,改善肝脂肪变性,但对肝脏病变的保护作用不如高浓度维生素C。
OBJECTIVE:Toexaminethepharmacologicalefectofβ-carboxyethylgermaniumsesquioxide(Ge-132)onscaveningfreeradicalandtheantioxidationinvivo.METHOD:Theefectsofthedrugsontheratsunderoxidativestressinducedbystrongwinewereinvestigatedbyobservingtheactivitiesofsuperoxidedismutase(SOD)inlivermitochondriaandpathologyofliverintherats.Wistarratswererandomlydividedintofourgroups(n=10):A:control;B:wine;C:wine+vitaminC;D:wine+Ge-132.Theratsinalgroupswerefedwithregularfood.Thedoseandrouteofadministrationofdrugsareasfolows:Strongwine(55%ethanol),5g/(kgd),igfrom8thdayfor2weeks.VitaminC200mg/(kgd)scfrom1stdayfor3weeks.Ge-132200mg/(kgd)igfrom1stdayfor3weeks.RESULTS:ActivityofCu,Zn-SODinlivermitochondriadecreasedinthestrongwinegroupcom-paredwiththecontrol(P<0.05),andtendedtoincreaseinthestrongwine+Ge-132groupandwine+vitaminCgroupcomparedwiththestrongwinegroup.Liversteatosiswaslessinthestrongwine+Ge-132groupthanthatinthestrongwinegroup,buttherewasnosignificantchangeinthestrongwine+vitaminCgroup.CONCLUSION:Ge-132couldprotectfromdecreasingtheactivityofCu,Zn-SODinlivermitochondriatoacertainextentandim-proveliversteatosisinstrongwinerat.TheefectsofGe-132inimprovingfatyliverwerenotbetterthanthoseofhighconcentrationvitaminC.
