摘要 目的研究人胎盘绒毛滋养层细胞中的10号染色体上缺失的磷酸酶和张力蛋白同源物基因(phosphatase and tensinhomology deleted on chromosome 10,PTEN)和增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)的表达与意义。方法PCR检测胎盘PTEN的外显子以证明PTEN基因的存在,原位杂交检测胎盘绒毛滋养层中PTEN mRNA的表达,免疫组化染色方法检测胎盘滋养层细胞中PTEN与PCNA的表达。结果在所分析的28例胎盘中,PCR均检测到PTEN基因的3,4,5外显子,93%(26/28)胎盘绒毛滋养层表达PTEN mRNA较强,同样地,86%(24/28)胎盘滋养层细胞中PTEN蛋白表达强,而胎盘绒毛内小的间质细胞不表达或弱表达PTEN mRNA和PTEN蛋白。PCNA主要在细胞滋养层细胞中表达强,在合体滋养层细胞中呈阴性。结论PTEN的表达与人胎盘绒毛滋养层细胞形成有密切的关系,细胞滋养层是胎盘绒毛中可增殖的细胞。 Objective To study the expression and significance of PTEN (phosphatase and tensin homology deleted on chromosome 10,PTEN)and PCNA (proliferating cell nuclear antigen,PCNA) in human placenta trophoblastic cells. Methods PCR was done to prove the existence of PTEN gene in trophoblastic cells of placenta. Expression of PTEN mRNA in trophoblastic cells was detected by in situ hybridization. Expression of PTEN protein and PCNA in trophoblastic cells were detected by immunohistochemistry. Results Exons-3,4,5 of PTEN gene were found by PCR in all of 15 placenta cases. Strong expression of PTEN mRNA was found in trophoblastic cells of 93 % (26/28 ) placenta cases. Likewise, strong expression of PTEN protein was found in trophoblastic cells of 86% (24/28) placenta cases. In the contrast, the small stromal cells in the placenta villus showed negative or weak PTEN mRNA and protein expression. Strong expres.sion of PCNA was mainly found in cytotrophoblast, the cells in syneytrophoblast showed negative. Conclusion Strong expression of PTEN has close relationship with the formation of trophoblastic cells in human placenta, the cells in cytotrophoblast have the ability to proliferate in placenta villi.
