体外培养的人牙髓细胞、牙周韧带细胞几种基质表达和碱性磷酸酶分泌的研究 被引量:5

摘要 目的:研究并比较培养的5~7代人牙髓细胞和牙周韧带细胞几种基质表达和碱性磷酸酶分泌的情况。方法:细胞培养、免疫组化、碱性磷酸酶活性分析。结果:两种细胞均在培养的第9天进入生长静止期,两种细胞培养液中碱性磷酸酶活性在增殖期达到高峰,以后活性下降。两种细胞均表达FN、Ⅲ型胶原、BMP。当培养的细胞密度较高时,FN和Ⅲ型胶原以纤维状结构覆盖于细胞表面。结论:传代的人牙髓细胞和牙周韧带细胞的上述几种基质表达方式、碱性磷酸酶活性相似。 Aim:To study and compare the expression of extracellular matrix proteins and alkaline phosphatase (ALP) activity in cultured human dental pulp cells and periodontal ligament cells. Methods:The cells′ culture,immunohistochemistry ,analysis of ALP activity in the supernatnant. Results:The cells reached stationary stage in the 9th day, ALP activity reached the maximum level in proliferating stage and then declined remarkably,FN.Ⅲcollagen and BMP were detected intracellularly .When the cells were cultured at high cell density ,the cells in the central area were covered by FN and type Ⅲ collagen fibrillar structures. Conclusions: The expressions of above extracellular matrix proteins and ALP activity in subcultured human dental pulps cells and periodontal ligament cells are similiar.
