摘要 小麦单细胞培养研究于1985~1995年进行,以六倍体普通小麦84—53的幼穗为外植体.在诱导胚性愈伤组织基础上.经振荡悬浮培养获得单细胞再生植株,建立体细胞无性系,并系统研究再生植株后代体细胞无性系的变异规律和遗传稳定性、体细胞无性系有利变异的选择及其在小麦品种改良中应用的可能性.本文报道有关单细胞培养和再生植株的研究结果. Studies were carried out from 1985 to 1995 at Northeast Agricultural University in Harbin. The procedueres of plant regeneration in single cell culture and somaclonal variation and screening were involved in the studies and the present report was the result concerning the single cell culture in vitro and plant regeneration. Young inflorescence of common wheat (Triticum aestivum L., line 84-53) was used as the explant in vitro culture to induce calli. MS medium was chosen as the basic medium and MD, MA and MF the modified. Fast growing and friable embryogenic calli in whiteor light-yellow color consisting of cells round in shape and thick cytoplasm were selected as the initial materials or suspension culture under the conditions of 25 / 18℃ (day/night), 120 ̄ 140 r/min and scattered light. Sub-culture was carried out twice a week. Single cells were collected by passing a series of nets with 40 ̄ 60 μmmeshes, then cultured under the same conditions as the initial suspension cultures except the day temperature 23℃ instead or 25℃. Semi-solid and doublelayer cultures were established when the suspension cells start of division. Calli that > 2 mm in diameter were transferred on MA or MF media, shoots and roots differentiated about one week later. Plantlers with threeroots were transferred into plastic pots. The first four regenerated plants belong to three cell lines were obtained on August 8 in 1989 and matured in February 1990 in greenhouse. The other 189 regenerated plants were obtained in 1990.
