摘要
目的检测138例非小细胞肺癌(NSCLC)组织中MMPs及其抑制剂TIMPs蛋白及mRNA水平的表达,探讨其与NSCLC生物学因素的关系。方法运用SP免疫组化技术检测MMP-2、MMP-9、TIMP-1、TIMP-3在138例NSCLC中的表达。RT-PCR检测14例NSCLC中MMP-9、TIMP-3的mRNA表达。结果NSCLC中MMPs和TIMPs的阳性表达率均显著高于正常肺组织及增生及不典型增生上皮,MMPs和TIMPs阳性表达率与TNM分期、淋巴结的转移有关,但与NSCLC分化程度、分类、肿瘤的大小无关;MMP-9、TIMP3的mRNA的表达与淋巴结转移相关;MMP-2的表达与NSCLC患者生存时间呈负相关,而TIMP3表达与生存时间呈正相关。结论MMPs和TIMPs与肺癌的浸润、转移有关,是NSCLC转移、TNM分期、预后的重要指标。
Objective To determine the expressions of MMPs and TIMPs and their eorrelation with biology factors in 138 human non - small eell lung carcinoma (NSCLC) patients at protein and mRNA level. Methods Immunobistochemistry S - P method was used to determine the expressions of MMPs and TIMPs in 138 NSCLC, and deteet the expressions of MMP9 and TIMP3 in 14 NSCLC tissues by RT- PCR. Results The positive expression rates of MMPs and TIMPs in NSCLC were higher signifieantly than normal tissue and hyperplastie or dysplastie epithelium; The positive expression rates of MMPs and TIMPs in NSCLC were associated with TNM stage,lymphatie node metastasis, but without association with eell differentiation of NSCLC and tumor types and tumor size. The expression of mRNA of MMP9 and TIMP3 was associated with lymph node metastasis. Negative eorrelafion was observed between the expression of MMP2 and survival time of NSCLC, but the expression of TIMP3 was positively associated with survival time. Conclusion MMPs and TIMPs are associated with infiltration and metastasis of lung eaneer and thus they are the important indexes for determination of lymph node metastasis, TNM stage and prognosis of lung eaneer.
