摘要
目的:观察牛磺酸对3T3细胞的增殖及其产生胶原和透明质酸的影响。方法:采用[3H]-胸腺嘧啶([3H]-TdR)、[3H]-脯氨酸([3H]-Pro)掺入和放射免疫分析法分别测定细胞增殖、胶原和透明质酸合成。结果:牛磺酸能显著抑制3T3细胞的[3H]-TdR和[3H]-Pro掺入,并呈时间、剂量依赖关系;对透明质酸的合成亦具有剂量依赖的抑制作用。此外,牛磺酸还明显提高细胞内cAMP水平。结论:牛磺酸能显著抑制3T3细胞增殖及胶原和透明质酸合成,其机理可能与促进cAMP生成有关。
To study the effects of taurine on proliferation and syntheses of collagen and production of hyaluronic acid of 3T3 cells. Methods: The cell proliferation and syntheses of collagen and hyaluronic acid were assessed by the incoporation of [3H]-TdR and [3H]-proline, and radioimmunoassay.Results: Addition of taurine(5~80 mmol/L)into culture medium significantly reduced [3H]-TdR and [3H]-proline incoporation in dose- and time-dependent manner, and decreased the hyaluronic acid level in dose-dependent manner. In additon, taurine markedly increased the intracellular cAMP level. Conclusion:Taurine can significantly inhibit the proliferation and syntheses of collagen and hyaluronic acid of 3T3 cells and cAMP formation-stimulating action might be one of the mechanisms involved in its inhibitory effect.
