栉孔扇贝C型凝集素基因的cDNA克隆和序列分析 被引量:2

摘要 利用本实验室已建立的栉孔扇贝cDNA文库中已知表达序列标签(expression sequence tag, EST),采用cDNA末端快速扩增(rapid amplification of cDNA end,RACE)技术获得目的基因的cDNA编码全序列。利用BLAST比对分析,结果发现目的基因序列与日本鳗鲡(Anguilla japonica)C型凝集素(C- type Lectin)E值达4e-19;利用Clustalw软件将目的基因与脊椎动物和节肢动物C型凝集素的多序列比对,发现克隆片段具有C型凝集素家族的标签序列模式且有很高的同源性,在该序列中形成二硫键的4个保守的半胱氨酸,与其他物种C型凝集素的二硫键结构非常相似;同时发现在其成熟肽中含有糖识别域(carbohydrate recognition domain,CRD),且Cys的位点是保守的。分析结果表明我们克隆的目的基因极可能为栉孔扇贝(Chlamys farreri)的C型凝集素基因。 This article describes the techniques of using rapid amplification of cDNA end RACE to obtain the whole cDNA code array for certain gene according to the known EST array of Chlamys farreri cDNA library, which has been established in lab. Comparing to the soft of BLAST, we found the E value of C - type Lectin of aimed gene array and Anguilla japonica is up to 4e - 19. Comparing the cloned aimed gene and C -type Lectin's poly array of vertebrate and arthropod using Chstalw soft, we found the matured peptide has carbohy6drate recognition domain, CRD, and the point of Cys is conservative. The cloned fragment has the same labelod array as C - type Lectin family and has the high homologous characteristic. According to the above analysis, the result comes out that the cloned aimed gene in this article may be the C - type Lectin gene of Chlamys farred.
