摘要
目的研究C60-氮芥对胃癌生长和转移的抑制作用。方法建立人胃癌裸鼠原位种植模型。40只荷瘤裸鼠随机分为4组,分别腹腔内注入生理盐水、C60、氮芥及C60-氮芥。观察原位种植肿瘤生长及转移情况,检测血清CEA值。结果生理盐水组、C60组、氮芥组和C60-氮芥组的抑瘤率分别为0%、4.6%、36.2%和41.4%;淋巴结转移率分别为90%、90%、70%和20%;肝转移率分别为80%、80%、60%和30%;血清CEA值分别为(32.6±4.5)ng/ml、(30.5±4.8)ng/ml、(17.9±4.1)ng/ml和(16.3±3.3)ng/ml。C60-氮芥组或氮芥组与生理盐水组比,对胃癌的生长的抑制作用差异有统计学意义,而C60-氮芥组与氮芥组比,对胃癌生长抑制作用差异不显著(P>0.05)。C60-氮芥组或氮芥组与生理盐水组比,对胃癌淋巴结转移生长后抑制作用差异均有统计学意义(P<0.05),且C60-氮芥组与氮芥组比,对胃癌的淋巴结转移生长的抑制作用差异有统计学意义(P<0.05)。结论C60-氮芥可抑制人胃癌裸鼠原位种植瘤的生长和转移,对淋巴结转移的抑制作用尤其明显。
Objective: To investigate the effects of C60-nitrogen mustard on the growth and metastasis of gastric cancer in nude mice. Methods:Forty male BALB/c nude mice were orthotopically transplanted with subcutaneous tumors and randomly divided into four groups as follows: NS group (n = 10), C60 group (n = 10) , nitrogen mustard group (n = 10) and C60-nitrogen mustard group (n=10). Different drugs were administered intrape,'itoneally. Mice were sacrificed at the 9^th" week "after operation. Implanted tumors and metastases were observed macroscopically and histologically. Serum CEA density was detected by using radio-immunity method. Results : Group C60-nitrogen mustard showed a statistically significant difference from the NS group in tumor growth (P 〈 0. 05 ). Compared to nitrogen mustard group, C60-nitrogen mustard group showed a statistically significant difference in lymph-node metastasis (P 〈 0. 05). The tumor inhibition rates of NS group, C60 group, nitrogen mustard group and C60-nitrogen mustard group were 0%, 4.6%, 36. 2% and 41.4%. The levels of serum CEA were 32. 6 ±4. 5ng/ml, 30. 5 ±4. 8ng/ml, 17.9 ±4. 1ng/ml and 16.3 ±3.3ng/ml. The rates of lymph-node metastases were 90%, 90%, 70% and 20%. The rates of liver metastases were 80%, 80% , 70% and 20%, Conclusion:C60-nitrogen mustard has inhibitory effects on growth and metastasis of human gastric tumor in nude mice, especially in lymph-node metastases.
