摘要
通过评价五指山小型猪(WZSP)近交系世代间的群体遗传结构和遗传变异的变化,为该品种早日用于实验动物研究提供可靠的理论依据。本试验先后采集五指山小型猪(WZSP)近交系F14-F16共50个个体DNA材料,利用不同染色体上的9个微卫星DNA标记对WZSP近交系群体中的3个家系进行了研究。试验结果证实,所检测的微卫星基因座有1-3个等位基因,各基因座等位基因频率在0.1250-1.0000之间,杂合度处于0.0000-0.6226之间,多态信息含量为0.0000-0.5480;平均杂合度为0.3849,平均多态信息含量为0.3270。3个家系在9个微卫星基因座的平均杂合度和平均多态信息含量分别为0.2934、0.2075、0.3005和0.2227、0.1588、0.2408。结果表明,WZSP已成为一个稳定的遗传群体,WZSP实验动物化培育已取得长足的进展。同时也从分子遗传学水平上证实WZSP近交繁育的可靠性,为WZSP近交系的开发利用提供了理论依据。
Genetic structure including diversity, polymorphism information content (PIC), heterozygosity (H) were chosen to study Wuzhishan miniature pig by using 9 microsatellite DNA markers. Microsatellite loci detected have 1 - 3 allelic genes, the frequence of allelic gene is between 0. 1250 and 1. 0000, heterozygosity is between 0. 0000 and 0. 6226, PIC is between 0. 0000 and 0. 5480, average heterozygosity is 0. 3849 and average polymorphism information content is 0. 3270. The average heterozygosity and polymorphism information content of three families are 0. 2934,0. 2075,0. 3005 and 0. 2227,0. 1588,0. 2408, respectively. The results indicated that with increasing of generation,genetic variation is reducing. It is suggested that WZSP is a genetically stability breed and satistifies the genetic standard of closed colony animal, and that the inbreeding degree of three lineages of WZSP are all high, and the richness of genetic diversity is lower than that in general commercial pig breeds. And this can provide theoretic foundation for the development of trial animals.
