摘要
目的观察HSPCO16重组蛋白对培养的低/高传代毛乳头细胞(DPC)的影响。方法消化对数期生长的第2代和第7代DPC,调整细胞浓度为3×105细胞/mL,接种于24孔细胞培养板;至对数生长期时加入HSPCO16重组蛋白,调整浓度为0.1mg/mL继续培养,倒置显微镜下定期观察;一组细胞采用流式细胞仪进行检测;另一组细胞内加入3H-TdR继续孵育6h,消化收集细胞,检测HSPCO16重组蛋白对DPC增殖的影响。结果HSPCO16重组蛋白处理的DPC增殖活跃,生长状态明显好于对照组;流式细胞仪分析和3H-TdR掺入试验均证实重组HSPCO16蛋白可促进第8代DPC的增殖及其DNA合成,而对第3代DPC则无明显作用。结论HSPCO16重组蛋白具有促进高传代DPC增殖的作用,可能是一种新的促进毛囊生长蛋白。
Objective To observe the effect of the recombinant HSPCO16 protein on low -/high passage dermal papilla cells(DPC). Methods To digest the passage 2 and 7 DPCs in log growth phase and to adjust the cell concentration to 3 ×10^5 cells/mL and incubate the DPCs to 24 hole cell culture flask. The recombinant HSPCO16 protein was added to DPCs in log growth phase and the concentration of protein was adjusted to 0. lmg/mL. The cells were observed by phase-contrast light electron microscopy. One group of cells was measured by flow cytometer,and the other group was supplemented with 3H-TdR incorporation and incubated for 6 hours. The cells were digested and the proliferative capacity was measured. Results The DPCs treated with recombinant HSPCO16 protein proliferated more actively than that of control DPCs. The results of flow cytometer and 3H-TdR incorporation showed that recombinant HSPCO16 protein promoted the proliferation and DNA synthesis of passage 8 DPCs. However, HSPCO16 protein had no effect on passage 3 DPCs. Conclusion Recombinant HSPCO16 protein had the effects of promoting the proliferation of high-passage DPCs and maybe a new protein to up - regulate the hair follicular growth.
