摘要
目的:研究1,25-二羟基维生素D3[1,25(OH)2D3]对大鼠脾辅助性T细胞(Th细胞)分化基因表达谱的影响。方法:采用近交系大鼠,随机分成实验组和对照组。实验组以1,25(OH)2D31μg/(只.d)连续灌胃14 d;对照组以同等体积赋形剂代替,第15天腹腔注射致死剂量脂多糖(LPS)10 mg/kg,6 h后处死,用功能分类Th1-Th2-Th3芯片检测脾Th细胞的基因表达谱。用Realtim e-PCR法验证差异表达的相关基因的结果。结果:功能分类Th1-Th2-Th3芯片共有99条基因,实验组与对照组对比,共有35条基因差异表达,占芯片基因总数的35.35%,其中12条基因上调,23条下调。差异表达的基因主要是细胞因子(受体)和调控Th细胞分化的转录因子。结论:1,25(OH)2D3抑制大鼠脾Th1型免疫反应,使免疫反应向Th2方向偏移。1,25(OH)2D3主要通过调节细胞因子基因和Th细胞分化信号通路中转录因子基因表达。
Objective:To investigate the effect of 1,25(OH)2D3 on gene expression profiles in Th cells ofratspleen. Methods : Lewis rats had been fed with l,25 ( OH ) 2D3 ( 1 μg per rat a day) for 14 days. The control group were administered the isotonic Na chloride as the same volume as the experimental group, and then LPS (10 mg/kg) was administered intraperitoneally in both groups at the 15th day,the animals were sacrificed 6 hours later. Gene expression in Th cells of rat spleen was analyzed by cDNA microarray technology. Results of gene array were verified by Real-time-PCR. Results:A total of 35 genes exhibited differential expression in Th cells of rat spleen compared with control. 12 genes were upregulated genes. and 23 genes were down regulated, which include Th cytokines and transcription regulators Conclusions:l, 25 (OH)2 D3 has a great influence on the gene expression in Th cells spleen. 1,25 (OH)2D3 inhibits Thl inflammation, bias the immune response towards Th2 pathway. (OH) 2D3 modulates Th cells differentiation regulating gene expression profiles of Th cytokines and of rat 1,25 transcription regulators.
