溶脲脲原体双相一步培养与PCR技术的临床应用

摘要 用溶脲脲原体(Ureaplasma urealyticum,Uu)双相一步培养法和PCR方法同时测定50例非淋球菌性尿道炎病人标本。双相一步培养法有24例阳性,阳性率为48%,PCR测定有20例阳性,阳性率40%,两者无显著差异(P>0.05)。24例培养阳性,PCR有5例阴性,26例培养阴性中,PCR有1例阳性。本文说明Uu双相一步培养法具有快速、准确、阳性率高,不易污染等优点,对于Uu的临床检测具有重要意义,PCR方法更快速简便,并直接能从样品中测定Uu的特异核酸片段,亦是Uu鉴定的有效手段,但不能取代培养法。 Double phase one step method and PCR were simultaneously used to detect Uu in samples from 50 cases of patients with non-gonococcal urethritis. The double phase one step method found 2.4 positive cases with a positive rate of 48% ,while PCR, 20 positive cases with a positive rate of 40%. There was no significant difference in positive rate finding between two methods. (P>0.05). Of 24 positive cases detected by double phase one step method, 5 cases were found to be negative by PCR. Of 26 negative cases detected by double phase one step method ,one case was found to he positive by PCR. Double phase one step method had advantages of faster speed,more accuracy,higher positive rate and less contamination ,it playing a significant role in clinical application for detection of Uu. Though PCR was even more simpler and faster and it could dected specific DNA sequance of Uu from samples directly,however ,it could not substitute double phase one step,method in clinical practice so far.
