辣椒离体培养再生植株的研究 被引量:9

摘要 以带叶柄子叶作为外植体,探讨了植物生长调节剂浓度及其组合、AgNO3、椰乳(CM)和基因型对辣椒分化形成不定芽以及不定芽伸长的影响,结果表明,MB(MS矿质元素+B5有机)培养基添加6-BA 5.0 mg/L和IAA 1.0 mg/L有利于辣椒子叶分化形成短缩不定芽,不定芽分化率达90.2%.将短缩不定芽转接到MB培养基添加6-BA 3.0mg/L、IAA 1.0 mg/L、GA32.0 mg/L、AgNO310.0 mg/L和φ=10.0%的CM培养基上,最高有40.0%的不定芽伸长.辣椒基因型不仅对不定芽的分化,而且对不定芽的伸长具有明显影响.伸长的不定芽培养在添加IAA 0.2 mg/L和NAA 0.1 mg/L MS培养基中,有85.0%的不定芽形成发达根系,移栽后全部正常开花结果. Effects of genotype, concerntration of plant growth regulator, AgNO3 and CM on adventitious bud induction of pepper were studied using cotyledon with petiole as explants. The results showed that the best differentiation medium for adventitious buds was MB ( MSmineral + B5 vitamin) + 6-BA 5.0 mg/L + IAA 1.0 mg/L and its differentiation rate was 90. 2%. The best elongation medium was MB + 6-BA 3.0 mg/L + IAA 1.0 mg/L + GA3 2. 0 mg/L + AgNO3 10. 0 mg/L + CM 10. 0% (φ) and its highest elongation rate was 40. 0%. Rooting medium was MS + IAA 0. 2 mg/L + NAA 0. I mg/L, and its average rooting rate was 85 %. All the plantlets flowered and set fruits normally in the field.
