地西他滨对MDS细胞株SKM-1的作用研究 被引量:7

摘要 目的:探讨去甲基化制剂地西他滨(5-aza-2’-deoxycytidine,Decitabine)对MDS-RAEB(骨髓增生异常综合征-原始细胞增多型)细胞株SKM-1株的影响以及机制。方法:用台酚蓝拒染法研究药物对SKM-1细胞生长曲线的影响,用NBT(硝基四氮唑蓝)还原试验和流式细胞技术研究药物对细胞的分化作用,用AnnexinⅤ-FITC标记了解药物对细胞的早期凋亡作用,用RT-PCR研究药物作用后P15INK4BmRNA表达的变化。结果:地西他滨对SKM-1细胞有生长抑制作用,能促进SKM-1细胞分化和诱导早期凋亡,上调细胞P15INK4BmRNA表达。结论:地西他滨可能通过上调P15INK4BmRNA表达而促进SKM-1株的分化和诱导细胞早期凋亡。
