摘要
目的:近年来许多研究表明骨髓间充质干细胞在基因治疗中可能作为基因运载细胞而发挥作用,本文就骨髓间充质干细胞作为基因运载细胞在基因治疗中的研究现状作一综述。资料来源:应用计算机检索Medline、Pubmed、Eldrvier、Ovid等数据库中1970-01/2005-12与骨髓间充质干细胞在基因治疗中研究进展相关的文章,检索词为"marrow mesenchymal stem cells,gene transfection,gene therapy",并限定文章语言种类为English。同时检索中文CNKI全文数据库2000-01/2006-06与骨髓间充质干细胞在基因治疗中研究进展相关的文章,检索词为"骨髓间充质干细胞,基因疗法,基因载体",并限定文章语言种类为中文。资料选择:纳入标准:①骨髓间充质干细胞的来源及生物学特性。②骨髓间充质干细胞作为基因运载细胞在基因治疗中的研究。排除标准:综述文献、Meta分析、重复研究。资料提炼:共收集到115篇有关骨髓间充质干细胞与基因治疗研究相关的文章,排除重复或类似的研究,30篇符合纳入标准。资料综合:骨髓间充质干细胞具有干细胞的共性,即自我更新及多向分化能力,而正是由于其容易体外扩增培养、易于外源基因的转染与表达、自体移植不易引起免疫排斥反应、体内移植后能迁移到病变部位等特性,使得骨髓间充质干细胞在基因治疗中引起人们越来越多的关注。利用骨髓间充质干细胞向肿瘤微环境趋向转移的特性,将人骨髓间充质干细胞作为细胞载体来开展的肿瘤基因治疗,取得了一定的治疗效果。但骨髓间充质干细胞的转染技术还存在很多问题。结论:骨髓间充质干细胞有希望作为一种较理想的基因运载细胞。
OBJECTIVE: Recant evidence suggests that marrow mesenchymal stem calls may be as a kind of useful delivery vehicles for gene therapy. This article is aimed to summarize the research progress of marrow mesenchymal stem calls as gene-delivery vehicles for gene therapy. DATA SOURCES:Using the key words of "marrow mesenchymal stem calls, gene transfection, gene therapy", we searched Medline, Pubmed, Eldrvier, Ovid databases for marrow mesenchymal stem caUs-releted articles and its study progress for gene therapy published between January 1970 and December 2005 in English. Meanwhile, We also searched the CNKI Full-text Database for the same articles published between January 2000 and June 2006, with the key words of "marrow mesenchymal stem cells, gene therapy, gene vector'in Chinese. STUDY SELECTION: Inclusive criteria were ①the source and biological characteristic of marrow mesenchymal stem calls and ②the study of bone marrow mesenchymal stem calls as gene-delivery vehicles for gene therapy. Exclusive criteria: review, Meta analysis and repeated articles were excluded. DATA EXTRACTION:Totally 115 articles about the study of bone marrow mesenchymal stem calls and gene therapy were collected, and repetitive or similar studies were excluded, and 30 met the inclusive criteria. DATA SYNTHESIS:Marrow mesenchymal stem calls had the multipotential as stem calls, such as self-renewal, multi-directional differentiation. Marrow mesenchymal stem calls for gene therapy had been paid more attention, because of easy amplification culture in vitro, transfection and expression of exogenous gene, no immunological rejection of autologous transplantation, transplanted into recipient might migrate into damage tissues or organs. Marrow mesenchymal stem calls as gene-delivery vehicles for gene therapy had obtained certain therapeutic efficacy by the characteristics of marrow mesenchymal stem calls transfer into tumor microenvironment. However, transfection technique of marrow mesenchymal stem calls existed many problems. CONCLUSION:Marrow mesenchymal stem cells are a promising target call for gene delivery.
