摘要
以玉米醇溶蛋白作底物,用Alcalase2.4FG碱性蛋白酶水解制备玉米抗氧化肽,SephadexG-25、DEAE-52纤维素柱层析纯化玉米抗氧化肽,建立小鼠急性酒精性肝损伤模型,研究其对小鼠急性酒精性肝损伤的保护作用。结果表明,制备玉米抗氧化肽的最佳酶解条件为:底物浓度为8%,[E]/[S]=2%,pH=9.0,时间3h,温度55℃。该条件下得到的玉米抗氧化肽对O2-·和·OH具有很强的清除能力,并对小鼠急性酒精性肝损伤具有一定的保护作用。氨基酸分析表明其富含Ala和Leu,且含有7种人类必需氨基酸,因此其具有很高的生物学活性和营养价值。
Corn anti-oxidative peptide was prepared by hydrolysising zein with Alcalase2.4FG alkaline protease, and was purified with SephadexG-25, DEAE-52 column chromatography. The hepatic injury models were induced by ethanol in mice, and the protective effects of corn anti-oxidative peptide on ethanol-induced acute hepatic injury in mice was investigated. The results showed that the optimum hydrolytic condition for preparation of corn antioxidative peptide were: [S]=8 %,[E]/[S]=2 %,PH=9.0, time 3 h,temperature 55 ℃.The corn antioxidative peptide prepared under this condition had higher scavenging activity to O2^-·and ·OH, and it had protective effects on ethanol-induced acute hepatic injury in mice. Amino acid composition of the corn antioxidative peptide was shown to be rich in Ala and Leu, and there were seven essential amino acids in it. So it had higher biological activity and nutritional value.
