摘要
目的探讨转化生长因子β1(TGF-β1)及其Ⅰ、Ⅱ型受体(TGF-βRⅠ、Ⅱ)在人类肾小球疾病中的肾内表达位点及其与各肾小球疾病的关系。方法采用免疫组化SABC法检测系膜增生性肾炎(MsPGN)22例,系膜毛细血管性肾炎(MPGN)6例,局灶节段性肾小球硬化(FGS)9例,膜性肾病(MN)4例中TGF-β1、TGF-βRⅠ、Ⅱ的表达。结果TGF-β1、TGF-βRⅠ、Ⅱ在各肾小球疾病中均有不同程度的阳性表达。各组中MPGN表达均呈强阳性,差异显著(P<0.05);各组球-管染色强度存在显著差异(P<0.05)。结论TGF-β1、TGF-βRⅠ、Ⅱ的过度表达参与肾小球疾病的进展;其在肾小球、小管及间质细胞均有表达位点,但在肾小管-间质表达呈强阳性,而肾小球呈弱表达。
Objective The aim of this study is to explore the expressions of transforming growth factor β1 (TGF- β1), and its types Ⅰ and Ⅱ receptors(TGF- β Type Ⅰ、Ⅱ Receptors, TGF- β1R Ⅰ、Ⅱ),and their roles and relationships in human glomerulonephritis.Methods Through immunohistochemistry, SABC, to assay the reflected changes in glomeru/ar expressions of TGF- β1 and TGF- β R Ⅰ and Ⅱ in selected biopsy samples of the following 41 disease cases: (MsP(3N) 22 cases; (MPGN) 6 cases, (FGS) 9 cases, (MN) 4 cases.Results Various degrees of significant immunoreactivity of TGF- β1, TGF- β R Ⅰ and Ⅱ were noted in the sampling disease cases. Amongst the samples, MPGN staining intensity was higher than the others in glomeruli (P〈0.05). Each sample group's tubulointerstitial immunostaining of TGF-β1, TGF- β R Ⅰ and Ⅱ all bore a significant difference (P〈0.05).Conclusion The excessive expressions of TGF- β1, TGF- β R Ⅰ and Ⅱ indicate their participations in the developments of human glomerulonephritis.These expressions were all noted in g/omeru/i and tubulointerstitial immunostaining, while more significant in tubulointerstitial and weaker in glomeruli.
