摘要
用ADP核糖多聚酶的竞争性抑制剂——3-氨基苯甲酰胺(3-aminobenza-mide,3AB)诱发中国仓鼠Wg3-h的姐妹染色体互换(SCE)。结果表明,3AB能间接地抑制DNA连接酶Ⅱ的活力,使BrdU(5-溴脱氧鸟嘧啶核苷)切除DNA后形成的单链断裂或连接延迟,导致SCE。用3AB处理BrdU加入后的第2个细胞周期,能增加诱发SCE的频率。
3-aminobenzamide (3AB), which is a competitive inhibiter of ADP-ribosonpolymerases was used in our experiments. It is a potent SCE inducer, and isnoncytotoxic, nonmutagenic and moreover. It does not inhibit the elongation ofDNA chain. The results showed that 3AB inhibited indirectly the activity of DNAligase II. It could delay the ligation of the single strand DNA after BrdU excision,and resulted in enhanced SCE levels. 3AB was more effective when it was introdu-ced during the second cell cycle after the addition of BrdU.
