摘要
以经过优选的山葡萄(Vitis amurensis Rupr.)幼嫩茎段为外植体,对适合植株再生的培养基激素配比、生根基质和生长环境进行了研究.结果表明:最优外植体诱导培养基为MS(改)+0.5 mg·L^-1BA+0.5 mg·L^-1KT+0.5 mg·L^-1NAA;诱导出的愈伤组织容易分化成茎丛,愈伤组织转接到MS(改)+0.5 mg·L^-1BA+0.1 mg·L^-1NAA内产生大量的不定芽;不定芽在MS(改)+0.05 mg·L^-1IBA的培养基内可诱导产生大量的根.生根试管苗驯化、移栽后,生根率最高的基质是苔藓,成活率达90%,而且在室外变温(温度12~30 ℃)的环境条件下生根最好.
The suitable medium, phytohormone mixture ratio and culture condition for plant regeneration were investigated using tender stem segments of Vitis amurensiss Rupr. as explants. Result showed that MS medium supplemented with 0.5 mg·L^-1 BA, 0.5 mg·L^-1 KT and 0.5 mg·L^-1 NAA could induce callus with high potential of organogenesis. The induced calluses subcultured to MS medium supplemented with 0.5 mg·L^-1 BA and 0.1 mg·L^-1 NAA produced a great deal of adventitious shoots. MS medium supplemented with 0.05 mg·L^-1 IBA was suitable for rooting of shoot. The rooting rate of seedlings grown in moss medium after acclimatization and transplantation reached 90 percent, and the higher rooting capability of seedlings was found in plastic tunnels at a changeable temperature of 12 Degree C to 30 Degree C.
