摘要
【目的】研究自体富含血小板血浆(PRP)和人重组骨形成蛋白(rhBMP-2)对下颌骨组织形成的促进作用。【方法】成年新西兰白兔45只,随机分为3组,每组15只,选择右侧下颌骨为实验侧、左侧下颌骨为对照侧。A组右侧下颌骨缺损区放置明胶海绵和PRP,B组右侧下颌骨缺损区放置明胶海绵和rhBMP-2,C组右侧下颌骨缺损区放置明胶海绵、PRP和rhBMP-2,各组左侧下颌骨缺损区只放置明胶海绵。分别于术后2、4、8周各取5只兔子进行放射性核素99mTc-MDP骨显像、X线片和组织学切片检查。【结果】A组动物右侧放射性核素计数在术后2、4周时明显高于左侧(Ρ<0.05);B组动物右侧放射性核素计数在第4周时高于左侧(Ρ<0.05),而在第2、8周时低于左侧(Ρ<0.05);C组动物右侧放射性核素计数在第2、4周时均高于左侧(Ρ<0.05),在第8周时两侧没有明显差别(Ρ>0.05)。【结论】PRP单独或与rhBMP-2复合均能促进下颌骨组织的增长。
[ Objective ] An animal study was carried out to investigate the influence of platelet-rich plasma (PRP) and recombinant human bone morphogenetic protein-2 (rhBMP-2) on the bone regeneration of mandibular defects in rabbits. [Methods] Forty-five New Zealand white rabbits were divided randomly into 3 groups: A, B, and C. All animals underwent general anesthesia and two 8 mm ×6 mm × 2 mm defects were drilled bilaterally in the mandibular bone. The right side defects were chosen as the experimental sides and implantation of glutin sponge with PRP was done in group A, rhBMP-2 in group B, and PRP and rhBMP-2 in group C, respectively. The left sides of defects were chosen as control side in which only the glutin sponge was implanted. The animals were killed after 2, 4, and 8 weeks. The specimens were observed by ^99mTc-MDP bone scanning, X-ray and histological examination. The results were compared between the experimental group and control group. [Results] At 2 and 4 weeks after surgery, the amount of ^99mTc-MDP on the right side was significantly higher than that on the left side in group A. In group B, the amount of ^99mTc-MDP of the right side was significantly higher than that on the left side at 4 weeks after surgery, but was significantly lower than that on the left side at 2 and 8 weeks after surgery. In group C, the amount of 99mTc-MDP on the right side was significantly higher than that on the left side at 2 and 4 weeks after surgery, and there was no significantly difference between the two sides at 8 weeks after surgery. [Conclusion] This study shows that the PRP alone or combine with rhBMP-2 can improve the generation of mandibular bone.
