摘要
目的:观察保肺定喘汤对慢性阻塞性肺疾病(COPD)大鼠血管内皮生长因子(VEGF)表达的影响。方法:模型组、保肺定喘汤预防组和治疗组采用慢性烟熏和气道内2次滴入脂多糖方法建立COPD大鼠模型。保肺定喘汤预防组和治疗组分别在造模前7天和造模第29天开始胃饲受试药物。采用免疫组化和逆转录-聚合酶链反应(RT-PCR)检测VEGF蛋白质和mRNA的表达。结果:造模3组具有COPD气道重构等病理改变。与正常对照组相比,模型组在中、小气道和肺动脉VEGF蛋白质表达显著增高;支气管肺组织VEGFmRNA表达也显著增高(P均<0.01)。保肺定喘汤预防和治疗两组,VEGF蛋白质和mRNA的表达则明显低于模型组(P<0.01-0.05)。结论:①VEGF在COPD大鼠气道重构中具有重要的作用。②中药保肺定喘汤可以减轻气道重构,其机制可能与调节VEGF在气道与肺动脉等的表达有关。
Objective : To observe the effects of Baofei Dingchuan Tang on the expression of VEGF in COPD model of rats. Methods: The rats of model group, prevention group and treatment group had been exposed in cigarette -smoking for 10 weeks, every rat was dropped LPS200p.g into airway on the first day and the 28th day . The prevention group were fed with 9.3g/kg of the Chinese medicine from the 7th day before molding, and the treatment group were fed with 9.3g/kg of medicine from 29th day after molding. To measure the area of VEGF expression in the bronchiole and pulmonary artery. To detect the expression of VEGF mRNA With RT - PCR. Results : There were remodeling of airway structure in three groups of COPD model of rats. Compared with the normal control group, the expression of VEGF in bronchiole and pulmonary artery of model group was higher significantly , so the expression of VEGF mRNA in lung tissue ( P 〈 0.01 ). The expression of VEGF in prevention and treatment groups were decreased significantly than that in model group ( P 〈 0.01 - 0.05 ) ,and so the expression of VEGF mRNA in lung tissue ( P 〈 0.01 ). Conclusion : (1)VEGF may plays an important role in airway remodeling process of COPD rats. (2)Baofei Dingchuan Tang can reduce the remodeling of airway by adjusting the expression of VEGF in airway and pulmonary artery.
