摘要
目的:分析周围静脉畸形组织中基质金属蛋白酶MMP-9的表达情况,研究其与周围静脉畸形临床分型的关系,初步探讨静脉畸形的发病机制。方法:对51例周围静脉畸形和10例正常微小静脉组织行免疫组织化学染色,观察并分析:①MMP-9及TIMP-1在周围静脉畸形与正常组织中的表达;②MMP-9与周围静脉畸形临床分型的关系;③MMP-9及TIMP-1在周围静脉畸形中表达的相关性。结果:MMP-9在周围静脉畸形中的表达率为82.35%,明显高于正常微小静脉组织(P=0.0002),在各分型中的表达有明显差异(P=0.0300),在有浸润与无浸润组的表达中有显著性差异(P=0.0043);TIMP-1在周围静脉畸形中的表达率为54.90%,与正常组织无明显差异(P=0.0842),与MMP-9的表达有差异(P=0.0145),但两者无明显正相关性(γs=0.3404)。结论:MMP-9在静脉畸形中的表达明显上调,在不同临床分型中的表达有明显差异,特别是在浸润型中呈明显高表达;MMP-9可作为静脉畸形临床诊断的参考指标,以及本研究临床分型的依据之一;MMP-9/TIMP-1平衡关系的破坏,可能是静脉畸形呈进行性生长的发病机制之一。
Objective To investigate the relationship between the expression of matrix metalloproteinase-9 (MMP- 9) and the sub-types of peripheral venous malformations. Methods MMP-9 and TIMP-1 expression were detected by immunohistochemical staining in 10 specimens of normal micro-veins and 51 specimens of peripheral venous malformation specimens. The relationship between the MMP-9 and TIMP-1 expression in venous malformations was analyzed. Results The expression rate of MMP-9 in venous malformation tissues was 82.35%, much higher than that of the normal microveins(P=0.0002). MMP-9 expression was significantly different among various sub-types of peripheral venous malformations (P=0.03); it was predominantly higher in subtypes with tissue invasion(P=0.0043).The expression rate of TIMP-1 in venous malformation tissues was 54.90%, which was similar to that in normal tissues(P=0.0842). No obvious relationship was found between MMP-9 and TIMP-1 expression (γa=0.3404). Conclusions MMP-9 is overexpressed in venous malformations, especially in sub-types with tissue invasion.Therefore, MMP-9 might be served as a diagnostic and sub-typing parameter of venous malformation. The disturbance of MMP-9 and TIMP-1 balance may lead to the progression of venous malformation.
