康莱特对HL-60/ADR细胞周期蛋白A表达的影响 被引量:1

摘要 目的探索康莱特对HL-60/ADR细胞周期蛋白CyclinA表达的影响。方法(1)康莱特作用HL-60/ADR细胞后,MTT法检查阿霉素对该细胞的抑制效应;(2)收集康莱特作用后的HL-60/ADR细胞,一定浓度阿霉素作用一定时间,流式细胞仪检查细胞内阿霉素的浓度;(3)康莱特作用对数生长期HL-60/ADR细胞,流式细胞仪分析细胞周期的变化及细胞周期蛋白CyclinA的表达。结果(1)1mg/ml康莱特作用HL-60/ADR细胞96h,阿霉素对该细胞的抑制效应较对康莱特未作用的HL-60/ADR细胞抑制效应强,P<0.05(;2)不同浓度的康莱特作用HL-60/ADR细胞96h后,1.5ug/ml的阿霉素作用6h,HL-60/ADR细胞内的阿霉素荧光强度均较对照组高,P<0.05,且具有剂量依赖关系;(3)1mg/ml康莱特作用HL-60/ADR细胞48h,HL-60/ADR细胞的细胞周期蛋白A的mean值(1207.9土109.6)较对照组(798.04土93.3)高,P<0.05。但康莱特对HL-60/ADR细胞周期的影响与对照组比较无显著性差异。结论康莱特具有上调HL-60/ADR细胞周期蛋白A的表达,此可能是康莱特增强HL-60/ADR对阿霉素敏感性的机理之一。 Objective To investigate the effect of Chinese Medicine Kanglaite on expression of cychnA in HL-60/ADR cells . Methods HL-60/ADR ceils at logarithmic growth phase were treated with Kanglaite , the inhibitory effect of Adrimay (ADR) on kanglaite-treated HL-60/ADR ceils were tested by MTT assay . The intracellular ADR fiourescence of certain concentration ADR-treated for certain time were tested by flow cytometry. The various phases of cell cycle and the expression level of cyclinA were detected by flow cytometry. Results (1) After 1mg/ml kamglaite treated HL-60/ADR cells for 96h,The inhibitory effect of ADR on the cells are statistically higher than that of no-kanglaite-treated groups,P〈0.05 .(2) After kanglaite treatment at different concentration ,The intrancellular ADR fiourescence of 1.5ug/ml ADR-treated for 6h are statistically higher than that of control group,P〈0.05.(3)1mg/ml Kanglaite treated HL-60/ADR for 48h, the expression of cyclinA was signiticantly increase.P〈0.05. Compared with the control group, the various phases of cell cycle were not significant. Conclusions Kanglaite could increase intracellular concentration of ADR and the expression level of cyclinA on HL-60/ADR cells so that the sensitivity of HL-60/ADR cells to ADR was increased significantly.
