酒精和高脂复合型小鼠肝损伤模型的建立

摘要 目的:建立脂肪堆积和慢性酒精性损伤复合型的小鼠肝损伤模型。方法:小鼠被随机分为模型组和对照组。模型组每天灌胃酒精,同时给予高脂饲料,对照组每天灌胃等量的水,给予正常饲料。灌胃4周后,采集被测试动物的肝脏,检测肝匀浆液的丙二醛(MDA)和谷胱甘肽(GSH)2项指标;采集被测试动物的血,检测谷丙转氨酶(ALT)、血清天门冬氨酸氨基转移酶(AST)和血清乳酸脱氢酶(LDH)3项指标。结果:灌胃4周后,模型动物组谷丙转氨酶(ALT)、血清天门冬氨酸氨基转移酶(AST)、血清乳酸脱氢酶(LDH)活力均显著增高,肝脏MDA指标升高(p<0.05),GSH显著降低(p<0.05)。结论:在灌胃酒精的同时饲喂小鼠高脂饲料,可以成功制备脂肪积累和慢性酒精损伤复合型小鼠肝损伤模型。 Objective: Estabhsh of alcohol and fat-induced liver injury mode in mouse. Methods: Selected mice were randomly divided into two groups, one was trial group and the other was control group. Optimized portions of alcohol and high-fat feed were administrated gastrically every day for trail group while the same volumes of water and normal feed were applied for control group. After 4 weeks, hvers and blood were collected from animals for 2 (MDA and GSH) and 3 (ALT, AST and LDH) assessments, respectively. Results: A significant increase in ALT, AST, LDH and MDA was observed while a reduction of GSH was seen. Conclusion: A supply of alcohol and high-fat feed by gastric administration is able to establish liver injury mode in mouse.
