摘要
目的观察大鼠肾小管间质纤维化过程中血清瘦素(leptin)水平的动态变化过程,探讨其在肾小管间质纤维化过程中的可能作用。方法32只雄性Wistar大鼠行单侧输尿管结扎术(UUO模型),造成结扎侧肾脏纤维化模型,于术后3d,7d,14d,21d各时点随机处死8只,另有8只行假手术作为对照组。收集血清测定其中瘦素及肌酐、尿素氮水平,肾组织切片行HE及Masson染色,供病理分析其纤维化程度。结果①血肌酐及尿素氮UUO组与假手术组相比无统计学差异;②UUO组肾小管间质损伤评分明显高于假手术组;③UUO大鼠较对照组血清瘦素水平显著升高,且在UUO各组之间血清瘦素水平随着肾纤维化程度的加重而增加。结论在肾小管间质纤维化过程中血清瘦素水平的增高与小管间质纤维化并存。
Objective To explore leptin expression in rat renal fibrosis induced by unilateral ureteral obstruction(UUO)and its role in the pathogenesis of renal fibrosis.Methods Forty male Wistar rats were randomly divided into sham operation group(n=8),3 d after UUO group(n=8),7 d after UUO group(n=8),14 d after UUO group(n=8),21 d after UUO group(n=8).The model rats were sacrificed at days 3,7,14,21 after UUO.The levels of serum leptin,urea nitrogen and creatinine at each time point were determined.Tissue sections were stained by HE and Masson staining.Results ①The levels of serum creatinine and urea nitrogen were not significantly different in UUO groups than in sham operation group.②The scores of renal interstitial lesion were significantly higher in UUO groups than in sham operation group.③The levels of serum leptin increased progressively at days 7,14 and 21 after UUO.Conclusion Leptin expression is enhanced in renal fibrosis,and may be involved in the pathogenesis of renal fibrosis.
