摘要
目的观察黏着斑激酶(FAK)与肝组织炎性反应及纤维化程度的关系及与血清HBV-DNA、HBeAg的关系。方法应用免疫组织化学方法测定64例慢性乙型肝炎患者肝组织中FAK、α-平滑肌肌动蛋白(α-SMA)的分布及表达,应用免疫荧光聚合酶链反应(PCR)方法检测血清HBV-DNA的含量,应用酶联免疫法检测血清HBeAg。结果(1)FAK在对照组中以阴性表达为主,随着肝纤维化及炎性反应程度的加重,FAK阳性细胞明显增多,与炎性反应及纤维化程度呈显著正相关,r值分别为0.712、0.730,P〈0.01。(2)α-SMA在对照组中以阴性表达为主。随着肝纤维化及炎性反应程度的加重,α-SMA阳性细胞亦明显增多,与炎性反应及纤维化程度呈显著正相关,r值分别为0.778、0.793,P〈0.01。(3)FAK与α-SMA呈显著正相关,r值为0.857,P〈0.01。(4)FAK、α-SMA阳性程度与血清HBV-DNA载量高低无明显关系,r值分别为-0.006、0.018,P〉0.05;与血清HBeAg阳性率高低亦无明显关系,r值分别为0.039、5.397,P〉0.05。结论慢性乙型肝炎患者,随着肝纤维化程度加重,肝组织FAK表达明显增加,与血清HBV-DNA、HBeAg无明显关系。
Objective To observe the relation between expression of focal adhesion kinase(FAK)and hepatic tissue inflammation,fibrosis degree,hepatitis B virus(HBV)-DNA and hepatitis B e antigen(HBeAg) in patients with chronic hepatitis B. Methods The liver biopsy was performed in 64 patients. The distribution of FAK and α-smooth muscle actin(α-SMA) in the liver tissues were assessed by mmunohistochemistry. HBV-DNA was detected by immunofluorescence polymerase chain reaction. HBeAg was detected by euzymelinked immunosorbent assay . Results FAK,α-SMA were mainly negative in normal livers,with the developing of liver fibrosis,the positive cells of FAK,α-SMA increased a lot. Both FAK and α-SMA correlated with inflammation and fibrosis grades and stages,r value was 0.712,0.730,0.778 as well as 0.793,respectively,P〈0.01. FAK correlated with α-SMA,rvalue was 0.857,P〈0.01. There were no significant relationship between FAK and serum HBV-DNA and positive rate of HBeAg,r value was -0.006 and 0.039,P〉0.05. There were no significant relationship between α-SMA and serum HBV-DNA and positive rate of HBeAg,r value was 0.018 and 5.397,P〉0.05. Conclusions FAK in liver tissues significantly increased with liver fibrosis stages and grades. There are no relationship between FAK and serum HBV-DNA and positive rate of HBeAg .
