人参皂甙Rg3诱导视网膜母细胞瘤HXO-RB_(44)细胞凋亡 被引量:17

摘要 目的观察人参皂甙Rg3对体外视网膜母细胞瘤(Rb)HXO-RB44细胞凋亡的作用。方法不同质量浓度的Rg3处理HXO-RB44细胞后,应用MTT比色法分析其细胞生长抑制作用。HOECHST33258染色法观察细胞的形态学改变,流式细胞仪测定48h细胞凋亡率。结果人参皂甙Rg3在一定范围内抑制体外培养的HXO-RB44细胞生长,其抑制率具有时间、质量浓度依赖性(P<0.01)。HOECHST染色可见实验组HXO-RB44细胞中致密强荧光。流式细胞仪检测凋亡率随药物质量浓度增加而增加。结论人参皂甙Rg3主要通过诱导HXO-RB44细胞凋亡达到抑制作用。 Objective Ginsenoside(Rg3) is an antitumor monomer extracted from ginseng. It can inhibit the growth of many tumor cells. The aim of this article was to investigate the action of Rg3 in the induction of retinoblastoma HXO-RB44 cells apoptosis in vitro and its possible mechanism. Methods HXO-RB44 cells were cultured in medium containing 10% fetal bovine serum. 10,25,50 μg/mL of Rg3 was added in medium respectively in experimental group, and the control group was set without Rg3. Effects of Rg3 on HXO-RB44 proliferation were evaluated by MTT colorimetric assay. The morphology change of HXO-RB44 cells was observed by HOECHST dyeing. The flow cytometry was used to detect the apoptotic rates of HXO-RB44 cells at 48 hours after Rg3 treated. Results The inhibiting rate of Rg3 on HXO-RB44 cells was gradually enhanced in 10,25,50 μg/mL of Rg3 addition group, showing a dose-dependent and time-dependent manner. HXO-RB44 cells showed the stronger fluorescence in Rg3 exposure group than control group. The nucleus of HXO-RB44 ceils became smaller and pyknic hyperfluorescence under the fluorescence microscope. The apoptotic rates of HXO-RB44 cells showed a increasing tendency in Rg3 exposure group with the enhance of Rg3 concentration( F = 16.97 ,P = 0. 000). Conclusion Rg3 can induce HXO-RB44 ceils apoptosis in vitro.
