摘要
目的:建立对蛇床子药材中花椒毒素、异虎耳草素、香柑内酯、欧前胡素、蛇床子素进行同时测定的RP-HPLC法。方法:采用Alltech C_(18)色谱柱(4.6mm×250mm,5μm),流动相乙腈-0.1%醋酸水溶液,梯度洗脱,流速:1mL·min^(-1),检测波长245,325nm;柱温为40℃。结果:花椒毒素、异虎耳草素、香柑内酯、欧前胡素、蛇床子素的线性范围分别为1~20μg·mL^(-1)(r=0.9999),1~20μg·mL^(-1)(r=0.9999),1~20μg·mL^(-1)(r=0.999 8),100~1200μg·mL^(-1)(r=0.9997),100~2000μg·mL^(-1)(r=0.9999),回收率均大于95%。结论:本法简便、准确、重复性好,适用于蛇床子中5种成分的同时测定。
Objective: To develop an RP-HPLC method for simultaneous determination of 5 constituents in Fructus Cnidii. Method: Analysis was performed on an Alhech C18 (4. 6 mm × 250 mm, 5 μm) column. The mobile phases were acetonitrile water and acetic acid with gradient elution. The flow rate was 1 mL · min^-1. The monitoring wavelength was 325 mn and 245 nm. The column temperature was 40℃. Result: The linear response ranges were 1-20 μg · mL^-1 ( r =0. 999 9) for xanthotoxin, 1-20 μg · mL^-1 ( r = 0. 999 9) for isopimpineUin, 11-20μg · mL^-1 ( r = 0. 999 8) for bergapten, 100-1 200μg · mL^-1 ( r = 0. 999 7 ) for imperatorin, 100-2 000μg · mL^-1 (r =0.999 9) for osthole. The average recoveries were all above 95%. Conclusion: The method is simple, sensitive and accurate with good reproducibility.
