摘要
目的:建立高效液相色谱-蒸发光散射法测定妥布霉素的含量。方法:采用AichromBon-AQC18(4.6mm×250mm,5μm);以0.2mol·L^-1的三氟醋酸为流动相;流速为0.6mL·min^-1;柱温为30℃;蒸发光散射检测器条件为漂移管温度110℃;栽气流速为2.0L·min^-1;撞击器模式:off模式。结果:妥布霉素在2.16~12.96μg范围内,进样量的对数与峰面积的对数呈良好的线性关系,r=0.9991。结论:本方法简便、快速、准确,可用于测定妥布霉素的含量。
OBJECTIVE To establish HPLC-ELSD(Evaporative Light Scattering Detector) method for the content determination of Tobramycin. METHODS The column: AichromBond-AQ C18 (4. 6 mm×250 mm, 5 μm) ;The mobiIe phase:0 2 mol·L^-1 trifluoroacetic acid (TFA) ; The flow rate: 0. 6 mL·min^-1 ; Evaporative llght-Scattering Detector(ELSD) : the temperature of the drift tube was set at 110 ℃,and the flow rate of gas at 2.0 L·min^-1 ;impact :off. RESULTS A double logarithmic calibration curve was obtained from 2. 16μ g to 12. 96 μg,r= 0. 999 1. CONCLUSION This method was simple,quick and accurate for the content determination of Tobramycin.
