摘要
目的采用柱前衍生法测定毛脉酸模根中氨基酸的含量。方法利用柱前衍生反相高效液相色谱(RP-HPLC)法测定毛脉酸模根中16种氨基酸。采用DikmaC_(18)色谱柱;流动相:A为0.05 mol/L乙酸钠溶液中含1%N,N-二甲基甲酰胺,B为乙腈-水(1:1),梯度洗脱;流速1.0mL/min;检测波长360nm;柱温40℃。结果氨基酸的摩尔浓度与峰面积成良好的线性关系,r均在0.999以上;16种氨基酸的回收率为94.7%~112.9%。结论该方法准确、重现性好,可用于毛脉酸模根中氨基酸的含量测定。
Objective To determine the amino acids in root of Rumex grnelini with pre-column derivatization. Methods Sixteen kinds of amino acids were determined in root of Rurnex grnelini by pre - column of RP-HPLC. HPLC analysis was performed on Dikma Cis column with mixture of 0.05mol/L sodium acetate to contain 1% N, N - dimethylformamide (A) and acetonitrile - wa- ter ( 1 : 1 ) (B) as mobile phase in gradient mode. The flow rate was 1.0ml/min. The detection wavelength was 360nm and the column temperature was 40℃. Results The molar concentrations of amino acids and peak areas achieved a nice linear relationship ( r was higher than 0. 999). The recoveries of sixteen kinds of amino acids were 94.7% -112.9%. Conclusion The method is accurate and repeatable for the determination of amino acids in Rurnex grnelini.
