摘要
采用显微注射的方法,将POMT/PGH、PSV40/PGH、POMT/PGH-MicrosatelliteDNA和PMHR等四种外源基因导入2592枚湖北白猪的受精卵,均获得了相应的转基因猪,基因导入的总效率为2.1%。对影响导入效率的几个因素进行了系统的实验。通过超数排卵,可以使供体母猪的获印数提高1.1倍;在异体移植的情况下,每头受体以移入20~29枚注射基因的胚胎为宜;采用自体移植,在移入胚数基本相同的情况下,其受孕率和产仔率比异体移植分别提高30.7%(P<0.05)和15.1%(P<0.01)。不仅提出了对转基因猪整合和表达检测的证据,而且提出了由目的基因决定的表型性状和传代方面的证据。
Four kinds of gene (such as POMT/PGH, PSV40/ PGH, POMT/PGH-Microsatellite DNA and PMHR) are microinjected into 2592 embryos of Hubei white pig, and the transgenic pigs have been obtained appropriately. Toeal transgenic efficiency is 2.1%. Some factors of effecting transgene have been tested for the system. By superovulation, the ovulation number is increased by 2.1 times. For every receptor,the implanted transgenic embryo number is 20-29. In planting the same number embryos, the pregnancy rate and pigging rate of pig with self-implant are higher than 30. 7% (p<0. 05) and 15.1% (P <0.01) approprately. The experiment brought the evidence about checking integration and expression of transgenic pig and phenotypic performances and genetic forms to be decided by aim gene.
