摘要
以3月龄高约25cm的海南龙血树为基本材料,以茎节纵切块为外植体,在改良MS+6-BA2.0mg/L+NAA0.1mg/L培养基中培养1个月后萌芽率为78.6%。萌芽在MS+6-BA1.5mg/L+NAA0.2mg/L培养基中连续两次继代培养,2个月后芽增殖倍数为2.9。丛芽及单芽都在MS+6-BA1.0mg/L+NAA0.2mg/L培养基中进行培养,1个月后丛芽的单芽增殖倍数为4.6,单芽增殖倍数为0.8。萌芽放入MS(改良)+NAA0.2mg/L培养基中进行培养,生根率为100%,移栽成活率为100%。
In this study,the three-month old Dracaena cambodiana about 25 cm high was used as the basic material. The tender stem fragments were cleaved lengthways into four pieces, each piece was used as the plantlet and induced in the medium modified MS supplemented with 6-BA2.0 mg/L and NAA0. 1 mg/L, the germination rate is 78% after a month culture. The proliferation multiple of the bud is 2.9 after the germination was continuously induced twice in the same medium MS supplemented with 6-BA1.5 mg/L and NAA0. 2 mg/L after two months,the medium was renovated once a month. What's more,the effect that the different form of bud used as a plantlet for subculture had on the proliferation multiple of the burgeon of Dracaena cambodiana when they were induced on the same medium has also been proved in this paper. Our result showed that the proliferation multiple of the clustering buds was 4. 6 and single bud 0. 8 when they were cultured on the same medium MS supplemented with 6-BA1. 0 mg/L and NAA0. 2 mg/L for a month. The rooting rate was 100% when the germination were cultured on the modified medium MS supplemented with NAA0.2 mg/L. And the survival rate of outplanting in nursery is 100%.
