摘要 目的:用MTT法检测树突状细胞(DC)杀伤肿瘤细胞的活性。方法:将人外周血分离出单个核细胞,在细胞因子的刺激下形成DC,观察其细胞形态、增殖情况,采用MTT法检测DC对肿瘤K562细胞的杀伤活性。结果:DC培养至第11天时,增殖高达120倍,经MTT法检测证实,DC对K562细胞的4h杀伤活性为31%。结论:MTT法检测进一步证实了DC对肿瘤细胞有较强的细胞毒作用。 Objective :To observe the anti-tumor effects of the dendritic cells(DCs) by MTT colorimetric assay. Methods: PBMCs were isolated from human peripheral blood by density centrifugation. Then DCs were generated from PBMC co-cultured with some cytokines. The cell mor- pholoy ,cell proliferative activity and anti-tumor effect were monitored. Results: It was detected that on the 11 th day of culture of DC, the pro- liferation of the cells increased by 120 times. MTT assay showed 31% of anti-tumor effect on K.562-cells by DC within 4 hours. Conclusion: The results suggest that in vitro the DCs might have a great quantity of proliferaion and high cytotoxicity against tumor,which can effectively kill the K562 cells.
