新型隐球菌逃避自然免疫与巨噬细胞产生NO关系探讨

摘要 【目的】探讨新型隐球菌感染与巨噬细胞产生NO的关系,并探讨TLR受体可能在其中所起的作用。【方法】用人工合成的非甲基化的CpG基序(TLR9配体)刺激巨噬细胞MH-S产生NO,在共培养物中加入新型隐球菌菌株,观察对NO合成的影响。【结果】经CpG-ODN刺激后,肺泡巨噬细胞株MH-S产生NO。由CpG-ODN诱导的NO合成可被加入的YC-11抑制。YC-11培养上清不能抑制巨噬细胞NO的合成。无荚膜新型隐球菌变异株也可抑制NO的产生。【结论】我们的实验结果显示,新型隐球菌可抑制巨噬细胞合成NO,该机制对新型隐球菌的预防和治疗有重要意义。证实了人工合成的非甲基化的CpG基序可刺激巨噬细胞合成NO。
