摘要
目的探讨子宫平滑肌瘤的发生与细胞凋亡的关系及抗单链DNA(single-strand DNA,ssDNA)抗体在检测子宫肌瘤细胞凋亡上的意义。方法采取免疫组织化学S-P法,用抗ssDNA抗体和Bax抗体检测40例子宫肌瘤组织和同一病例正常子宫肌组织中ssDNA的产生状况和bax基因的表达情况。结果Bax和ssDNA在正常子宫肌组织中免疫染色阳性率和免疫染色强度均显著高于肌瘤组织,两者比较,差异有显著意义(P<0.01)。在子宫平滑肌中,Bax蛋白表达量与ssDNA产生量之间存在着正相关(r=0.720,P=0.000)。结论Bax表达的下调,可能在子宫肌瘤的发生过程中起一定的作用;使用抗ssDNA抗体的免疫染色方法,可以准确检测凋亡细胞。
Objective To approach the association of the uterine leiomyoma genesis with cell apoptosis as well as the significance detecting the cell apoptosis in uterine leiomyoma by anti single-strand DNA (ssDNA)antibody. Methods Immuno histochemical S-P staining was used to test ssDNA and the protein expression of bax gene in both uterine leiomyoma and normal myometria adjacent to leiomyoma of 40 cases with uterine leiomyoma by anti-ssDNA antibody and anti Bax monoantibody. Results The positive rates and intensity of immunostaining both ssDNA and Bax were much more higher in the normal myometria than those in the uterine leiomyoma (P〈0.01), and there was the positive correlation between the quantity of Bax protein expression and the amount of the ssDNA (r= 0. 720, P = 0. 000). Conclusion Down-regulation of Bax expression may play an important role in genesis of the uterine leiomyoma, apoptosis cells in uterine smooth muscle could be detected exactly by anti-ssDNA antibody.
